CITE-seq, cell hashing and 10x Feature Barcoding all rely on the same reagent: an antibody labeled with a barcoded oligo, known as an antibody-derived tag (ADT). For many studies, the ready-made version has two problems.
The clone you need isn't in the catalog. Ready-made panels come with a fixed list of markers and clones. If your target calls for a different clone, or an antibody you have already validated elsewhere, the fixed panel can't supply it.
Conjugating it yourself is hard to keep consistent. The number of oligos on each antibody affects the signal, and both too few and too many compromise it. Getting the degree of labeling right, and repeating it across a whole panel, is difficult to do reliably by hand.
Under- or over-labeling an antibody with oligos can cost you a marker across the entire run.
