What is a custom PLA probe pair?
A proximity ligation assay detects two proteins that sit close together by conjugating each detection antibody to a short oligonucleotide. When the two probes are in proximity the oligos are ligated and amplified into a bright, countable signal. We conjugate the complementary oligos directly onto your chosen primary antibodies, so the pair is ready to run without species-matched secondary probes.
How is this different from off-the-shelf Duolink probes?
Catalog PLA kits rely on anti-species secondary probes, which means your two primaries must come from different host species and must be species the kit supports. Conjugating the oligos directly onto your primaries removes both limits: you can pair same-species clones, use any host, and detect targets that have no catalog probe. It is a flexible alternative when the standard kit does not fit your pairing.
Can you pair two antibodies from the same species?
Yes. Because the oligos go directly onto each primary, host species no longer has to be different. Two rabbit monoclonals can form a working PLA pair, which is not possible with secondary-based PLA.
How small a starting amount can you work from?
From just 20 µg of antibody per probe, so you can validate a pairing without committing precious primary stock before you know it works.
